    Human Monoclonal Antibody Against West Nile Virus Shows Promise in Animal Studies

    Leiden, The Netherlands, June 10, 2005 - Dutch biotechnology company Crucell N.V. (Euronext, NASDAQ: CRXL) today announces the discovery of a monoclonal antibody for protection against West Nile virus. Study results have demonstrated that the fully human monoclonal antibody, derived from patients who survived West Nile infection, effectively neutralized the virus in cell culture and fully protected mice from an otherwise lethal infection.

    The Director of Crucell's Antibody Discovery Group, Dr Jan ter Meulen, makes the announcement today at the 8th Annual KIT (Congress for Infectious Diseases and Tropical Medicine) conference in Hamburg, Germany. The discovery of the West Nile antibody follows the Group's discovery of antibodies against SARS and Rabies during 2004. It also complements Crucell's West Nile virus vaccine development program that is due to enter clinical trials at the end of 2005.

    While the vaccine will be targeted at preventing infection in the elderly population most at risk of developing the most severe form of the disease, the antibody may be applied post-infection in specific cases to prevent the potentially fatal onset of encephalitis.

    "I'm very proud that we have delivered another early-stage antibody against a serious infectious disease, as promised at our analyst day last November," said Crucell's Chief Scientific Officer Jaap Goudsmit. "West Nile virus continues to cause serious annual epidemics in the United States and elsewhere. While our determined efforts to develop a vaccine continue, an antibody may provide another route to prophylaxis, providing passive immunity in the event of an outbreak, and possibly therapy for patients who might otherwise develop encephalitis."

    Since its emergence in New York in 1999, West Nile virus has spread throughout the entire United States. Primarily transmitted through the bite of infected mosquitoes, it can cause a fatal form of encephalitis, or inflammation of the brain, in humans and some animals. Health authorities in the U.S. now view the disease as a recurring threat, surfacing every summer with warm and humid weather. In 2004, 2,470 recorded cases claimed 88 lives.

    Menselijke monoklonale antistof tegen West-Nijlvirus lijkt veelbelovend in dierproeven

    Leiden, 10 juni 2005 - Het Nederlandse biotechnologiebedrijf Crucell N.V. (Euronext, Nasdaq: CRXL) maakt bekend dat het een monoklonale antistof heeft ontdekt die bescherming biedt tegen het West-Nijlvirus. De onderzoeksresultaten tonen aan dat deze volledig menselijke monoklonale antistof, die is geïsoleerd uit patiënten die een besmetting met het West-Nijlvirus hebben overleefd, het virus effectief neutraliseerde in een celcultuur en muizen volledig beschermde tegen een besmetting die anders dodelijk zou zijn geweest.

    Het hoofd van Crucell's Antibody Discovery Group dr. Jan ter Meulen maakt de ontdekking vandaag bekend op het 8ste jaarlijkse KIT-congres (Congres voor Infectieziekten en Tropische Geneeskunde) in Hamburg. Voor de ontdekking van deze antistof tegen het West-Nijlvirus had de groep in 2004 ook al antistoffen tegen SARS en hondsdolheid ontdekt. De antistof vormt een goede aanvulling op Crucell's ontwikkelingsprogramma voor een vaccin tegen het West-Nijlvirus, in het kader waarvan klinisch onderzoek nog voor het eind van 2005 van start zal gaan.

    Terwijl het vaccin is bedoeld om infectie te voorkomen bij ouderen, die het grootste risico lopen om de meest ernstige vorm van de ziekte te krijgen, kan de antistof worden gebruikt bij mensen die al besmet zijn, om te voorkomen dat bij hen een mogelijk dodelijke hersenvliesontsteking optreedt.

    "Ik ben er trots op dat wij er opnieuw in geslaagd zijn een antistof voor een ernstige infectieziekte te isoleren, zij het dat we nog maar aan het begin van het onderzoek staan. Hiermee maken wij waar wat wij beloofd hebben op een bijeenkomst met beleggingsanalisten eind november vorig jaar," zei Jaap Goudsmit, Chief Scientific Officer van Crucell. "Het West-Nijlvirus veroorzaakt ieder jaar ernstige epidemieën in de Verenigde Staten en elders. Wij blijven natuurlijk werken aan het ontwikkelen van een vaccin, maar het hebben van een antistof is een andere manier om ervoor te zorgen dat mensen niet ziek worden. Als er een epidemie uitbreekt, biedt een antistof passieve immuniteit en kan het als medicijn worden gebruikt voor mensen die anders mogelijk hersenvliesontsteking zouden krijgen."

    Sinds het in 1999 voor het eerst in New York werd gesignaleerd, heeft het West-Nijlvirus zich verspreid over de gehele Verenigde Staten. Het wordt voornamelijk verspreid door de beet van geïnfecteerde muggen en kan een dodelijke vorm van encefalitis - dit is hersenvliesontsteking - veroorzaken bij de mens en bij sommige dieren. De gezondheidsautoriteiten in de Verenigde Staten beschouwen de ziekte nu als een weerkerende bedreiging, die elke zomer bij heet en vochtig weer de kop opsteekt. In 2004 werden er 2470 gevallen van de ziekte gemeld, waarvan 88 met dodelijke afloop.

    Misschien een stomme vraag, maar is het niet een beetje dubbel op om zowel een vaccin als een antistof te ontwikkelen? Als je een goedwerkend vaccin hebt (en iedereen daar mee ingeend wordt) wordt een antistof toch overbodig? Het bestaan van een antistof zal de vraag naar vaccin misschien doen afnemen. Een antistof ontwikkelen waar nog geen vaccin voor bestaat/in ontwikkeling is, zal toch een stuk interessanter zijn of niet?

    Is geen stomme vraag. Zie ook mijn opmerking in BioStefans draadje.

    Een vaccin is actieve vaccinatie waarbij het een tijdje kan duren voordat er weerstand is opgebouwd.
    Antilichamen kunnen na toediening eventueel direct bescherming bieden en daarom zouden de doelgroepen wel eens anders kunnen zijn.

    Een andere mogelijkheid is dat het WNV vaccin met name T cel immuniteit oproept, terwijl antilichamen door de B cellen worden gemaakt. Op die manier zouden ze complementair werken.
