   Crucell Announces Collaboration with GlaxoSmithKline Biologicals on Second Generation Malaria Vaccine Candidate
   
   Leiden, the Netherlands (April 6, 2010) - Dutch biopharmaceutical company Crucell N.V. (NYSE Euronext, NASDAQ: CRXL; Swiss Exchange: CRX) today announced that it has signed a binding letter of agreement with GlaxoSmithKline Biologicals (GSK) to collaborate on developing a second generation malaria vaccine candidate. Pre-clinical data from earlier studies indicated significantly enhanced immune responses against the malaria parasite (circumsporozoite stage of the Plasmodium falciparum) when Crucell's Adenovirus (AdVac®) technology and GSK's RTS,S/AS technology are used in combination, versus either component alone.
   
   
   
   Under the terms of the letter of agreement, Crucell will contribute its recombinant malaria vaccine candidate, Ad35-CS, based on Crucell's AdVac® technology and PER.C6® manufacturing platform and GSK will contribute its late stage malaria vaccine candidate RTS,S/AS. Financial details of the agreement were not disclosed.
   
   
   
   In October 2003 Crucell announced a program with the Walter Reed Army Institute of Research and GSK. The parties entered into a Cooperative Research and Development Agreement (CRADA) to evaluate Crucell's malaria vaccine candidate in pre-clinical studies. Crucell's vaccine candidate was tested as a stand-alone and in combination with GSK's malaria vaccine candidate RTS,S/AS. The tests conducted were designed to assess whether a combination of the GSK vaccine candidate with Crucell's vaccine candidate could lead to improved results. These pre-clinical studies showed significant enhancement of the immunological response when both vaccines were used in combination[1].
   
   
   
   This collaboration now intends to progress this concept into human clinical studies. The companies will seek third party funding to advance the Ad35-CS and RTS,S/AS prime-boost candidate into a Phase I/IIa clinical challenge trial in the United States. Pending the results of the Phase I/IIa trial, the companies expect to advance the prime boost candidate in further clinical studies with the support of public or non-profit partners who are interested in accelerating the development of a malaria vaccine.

   AdVac® technology is a vaccine technology developed by Crucell and is considered to play an important role in the fight against emerging and reemerging infectious diseases, and in biodefense. The technology supports the practice of inserting genetic material from the disease-causing virus or parasite into a 'vehicle' called a vector, which then delivers the immunogenic material directly to the immune system. Most vectors are based on an adenovirus, such as the virus that causes the common cold.
   
   The AdVac® technology is specifically designed to manage the problem of preexisting immunity in humans against the most commonly used recombinant vaccine vector, adenovirus serotype 5 (Ad5), without compromising large-scale production capabilities or the immunogenic properties of Ad5.    About Crucell's malaria vaccine

   @king1: realiseer je wel dat het nog heel lang gaat duren voor alle klinische fasen zijn doorlopen, met het bijbehorende afbreukrisico. Ook het feit dat er zwaar zal worden geleund op financiering door overheden en non-profits zal vermoedelijk t.z.t. consequenties hebben voor de prijs (wat voor de mensheid overigens een goede zaak is).
   
   Maar deze samenwerking staat in ieder geval prima op het lijstje van partnerships en sluit perfect aan op de missie van Crucell.
   
   Maar de kassa zal dus harder gaan rinkelen van de recente QV-ontwikkelingen.

    Het enthousiasme straalt er niet bepaald vanaf. Meer wantrouwen, wellicht met Flucell in gedachten:
    
    Het is 'binding', dus GSK kan niet een van de kansrijkste concurrent-malaria-vaccins een zachte dood laten sterven door middel van een 'samenwerking'.
    Welke details Crucell in het agreement heeft laten opnemen om dat te voorkomen weten we niet, maar hopenlijk telt een gewaarschuwd Crucell voor twee.
    
    De taal van't PB bevestigt, net als de lange tijd die de totstandkoming van het agreement heeft vereist, dat dit (wellicht net als bij Rabies deal) een verstandshuwelijk is:
    Wetenschappelijk is het zeer duidelijk dat de combinatie RTS,S en Ad-35 de best mogelijke combinatie is voor een effectief vaccin. Dit dwingt de twee onwillige partijen tot samenwerking, wellicht onder enige druk van externe financiers (Bill Gates c.s.).
    
    Het belangrijkste is natuurlijk dat beide partijen nu eindelijk officieel weer op lijn zitten, en dat daarmee een effectief vaccin dichterbijkomt. En, belangrijk voor Crucell zelf, de kans op een vaccin met een Crucell component erin is een stuk groter geworden.
    
    Koersreactie zal wel tegenvallen, omdat 't natuurlijk geen echt nieuws meer is. En toen het nog wel echt nieuws was (2009 results), werd het overschaduwd door andere zaken.

    AMSTERDAM (Dow Jones)--De samenwerking met GlaxoSmithKline voor een malariavaccin is een positieve ontwikkeling voor Crucell, reageert SNS Securities. Vanwege het uitblijven van financiele details en de studies die het middel nog moet ondergaan verwacht de analist evenwel beperkte invloed op het aandeel.     Bij Crucell houden kopers en verkopers elkaar in balans. Dit vertaalt zich op de grafiek in een zijwaarts gericht koersverloop tussen steun 15,05 (gevormd op 12 maart) en weerstand 15,70 (top van 17 maart). We wachten op nieuwe technische signalen.

    Wat wordt nu de volgende stap?
    
    Nu Crucell's Ad35 phase I eindelijk is afgerond, is het tijd voor de volgende stap. Als alle gegevens zijn ge-analyseerd en Ad35 blijkt veilig, kan volgens mij nu meteen worden doorgestoten naar een RTS,s prime/Ad35 boost phase II trial, i.p.v. eerst een go-it-alone Ad35 phase II, terwijl uiteindelijk toch voor de combinatie wordt gekozen.
    
    Ik denk/hoop niet dat Ad35 en RTS,s, die nu allebei apart een phase I hebben doorlopen, ook nog eens samen eerst een phase I doormoeten.
