    Op verzoek hier wat info die ik verzameld heb over SIRNA ticker RNAI. Dit draadje is bedoeld om snel iets te kunnen vinden zodat je niet draadjes als "OOK SIRNA" helemaal hoeft door te spitten, toevoegen van links, artikelen, persberichten enzo zijn zeer welkom.

    SIRNA ontwikkeld RNA technologie waar afgelopen jaar de eerste doorbraken zijn geboekt tegen huntingtons disease en recent tegen hepatitis, beide in muizen overigens. Vandaar ook recent een koersexplosie. De komende jaren kunnen zeer interessant zijn als er goede veredere resultaten geboekt worden aangezien deze techniek als een nieuwe type medicijn wordt gezien.

    Nieuws! Ik zat gisteren nog te twijfelen of ik niet al te vroeg was ingestapt. Dit is toch wel een verklaring voor alle insider buying en de koersstijging de afgelopen maand.

    Sirna Therapeutics, Inc. (Nasdaq: RNAI - News) announced today the publication in Nature Biotechnology of a breakthrough study demonstrating a 95% knockdown of hepatitis B virus (HBV). In the study, Sirna used its chemically optimized short interfering RNA (siRNA) at 1, 3 and 5 mg/kg together with encapsulation and delivery technology provided by Protiva Biotherapeutics Inc. This is the first demonstration of systemic siRNA efficacy at therapeutically relevant doses and establishes a strong scientific foundation for broad human application of RNAi-based therapeutics.

    The study demonstrated that Sirna's chemically optimized and encapsulated siRNAs have high potency and prolonged duration of activity in vivo. The siRNA formulation, targeting HBV, was administered by standard intravenous injection to mice carrying replicating HBV. Three therapeutically relevant doses of 1, 3, or 5 mg/kg resulted in up to a 95% dose dependent reduction in serum HBV DNA levels. A similar reduction was observed in Hepatitis B Surface Antigen protein levels. The anti-viral activity persisted for at least seven days.

    This unprecedented efficacy correlates with a significantly longer circulation time of siRNAs in blood plasma and residence time in the liver compared to unmodified and non-encapsulated siRNAs. Importantly, Sirna modified and optimized the siRNAs used in the study to abrogate the induction of serum interferon-alpha (IFN-alpha) and inflammatory cytokines (IL-6, TNF-alpha) thereby further demonstrating Sirna's capability to chemically modify RNAi-based therapeutics to modulate cellular responses.

"With these results, we have established a new benchmark for siRNA efficacy in vivo. This is the second study published by Sirna scientists that validates the efficacy of chemically optimized siRNAs in vivo. These robust results further underscore Sirna's leadership position and extensive experience in RNA biology and chemistry. Sirna is building on these landmark results as we focus our human clinical program on hepatitis C. We continue to improve the efficacy of our anti-viral compounds through proprietary modification and delivery approaches and plan to initiate hepatitis C clinical trials next year."

    Ian MacLachlan, PhD, Chief Scientific Officer of Protiva Biotherapeutics Inc., said, "The high degree of efficacy observed in this study is superior to all other previously reported data for any systemically delivered siRNA and is a direct result of combining Sirna's expertise in chemical modifications with Protiva's encapsulation and delivery technology."

    Sirna initiated primate studies in its hepatitis C program in June 2005 and expects to commence Phase I human clinical trials in 2006.

    Sirna Therapeutics and Allergan Enter Into Strategic Ophthalmology Alliance

    SAN FRANCISCO, Sept. 29 /PRNewswire-FirstCall/ -- Sirna Therapeutics, Inc. (Nasdaq: RNAI - News), announced today it has entered into a multi-year alliance with Allergan, Inc. (NYSE: AGN - News) to develop Sirna-027, a novel RNAi-based therapeutic currently in Phase I for age-related macular degeneration (AMD), and to discover and develop other novel RNAi-based therapeutics against select gene targets in ophthalmic diseases. Sirna will receive an initial payment of $5 million and be eligible for development milestones of up to $245 million in addition to research funding and royalties on the worldwide sales of products resulting from the alliance. Sirna also will receive contract manufacturing revenues.


    "We are pleased to enter into this exciting collaboration with Sirna, a pioneer in RNAi-based therapeutics," said Dr. Scott Whitcup, Allergan's Executive Vice President, Research & Development.     For the quarter ended September 30, 2005, Sirna reported a net loss applicable to common stock of $4.2 million, or $0.08 per share, compared to a net loss applicable to common stock of $7.8 million, or $0.21 per share, for the same period in 2004. Sirna ended the third quarter with $44.7 million in cash and marketable securities.

    Revenues for the third quarter of 2005 were $1.9 million, up from $84,000 for the same quarter in 2004. The increase is due to a $1.8 million success milestone payment received in July 2005, as part of Sirna's oncology collaboration with Eli Lilly and Company. Subsequent to the third quarter, in October, the Company received a $5 million upfront payment from Allergan, Inc., to initiate a strategic alliance in ocular diseases.

    Operating expenses decreased to $6.3 million for the third quarter of 2005 compared to $7.9 million for the third quarter of 2004. The decrease is primarily due to lower operating costs and lower license fees as compared to the third quarter of 2004.

    In July and August, Sirna closed a $28 million private placement issuing 17,506,250 shares of common stock and 6,302,246 warrant shares of common stock. Sirna expects that, together with existing cash balances, the financing provides the Company in excess of two years operating cash. These funds should enable the company to continue advancing its hepatitis and dermatology compounds with the goal of filing Investigational New Drug applications with the FDA for both programs in 2006.

    een korte uitleg over RNA:
    Zoals de naam laat veronderstellen, is de interferentie van RNA (RNAi) een cellulair mechanisme om de expressie van genen en de replicatie van virussen te regelen. RNAi technologie is een betrekkelijk recente ontdekking die door wetenschappers wordt verondersteld een belangrijk aspect van het natuurlijke defensiemechanisme te vormen van een cel tegen parasitische virussen.

    Door middel van deze nieuwe technologie probeert Sirna Therapeutics medicijnen te ontwikkelen tegen o.a. Hepatitis c, astma, Huntington’s Disease en AMD (Age related Mascular Degeneration). Onlangs zijn er positieve resultaten geboekt tegen ziektes als Huntington’s Disease en Hepatitis C, weliswaar in muizen. Dit heeft gezorgd voor een enorme koersexplosie. Verder heeft het bedrijf heeft voor de verschillende onderzoeken diverse partners binnengehaald, waaronder Eli Lilly, Targeted Genetics, Archemix Corporation, Protiva Biotherapeutics en Allergan. Deze laatste zal Sirna Therapeutics $ 5 mln betalen in haar verdere ontwikkeling van Sirna-027 (een produkt dat ontwikkeld wordt tegen AMD) en zal bij een uiteindelijke behandeling van AMD d.m.v. het gebruik van Sirna-027 tot en met $ 245 mln in mijlpaal bedragen betalen.

    Ondanks een veel belovende pipeline, positieve resultaten en nieuwe partners lukt het het aandeel toch om onderuit te gaan op de Nasdaq. We pakker er voor het gemak een grafiek bij:

    25 juli 2005 maakt Sirna Therapeutics positieve studie resultaten met betrekking tot hepatitis B virus. Het aandeel stijgt in 1 dag $ 1.42 en sluit op $ 4.04. De dagen erna stijgt het aandeel naar nieuwe year highs.
    10 augustus 2005 maakt Sirna Therapeutics cijfers bekend. Niets extreems voor een biotech bedrijf (gelet op de cashburn), maar genoeg aanleiding om het aandeel onderuit te halen. Het aandeel keldert in de dagen erna tot een dieptepunt van $ 3.10.
    Dagen na de cijfers herstelt de koers langzaam boven de $ 4.00. Op 29 september maakt Sirna Therapeutics Allergan als nieuwe partner bekend. Het aandeel vliegt ver boven de $ 5.00 (intraday high $ 5,62) om vervolgens onder de $ 4.60 te sluiten.
    Nu zijn we hier beland. Het aandeel is sinds de nieuwe partner bekendmaking verder gedaald en noteert op moment van schrijven $ 3.15.
    In iets minder dan een maand van $ 5.60 naar $3.12 vinden wij van Pennystock Express iets teveel van het goede. Gelet op de omzetten die zijn gehaald bij de enorme stijgingen kun je stellen dat de meeste mensen een gemiddelde hebben ver boven de $ 3.50. Het lijkt erop dat er flink geshort wordt in het aandeel, dat verklaart misschien ook de plotselinge heftige stijgingen. De vraag is natuurlijk hoever durven de shorters te gaan. Er is een lichte steun rond de $ 3.00. Wij zijn van mening dat het aandeel op dit moment ondergewaardeerd en oversold is en beschouwen de huidige koers als zeer interessant. Natuurlijk is het mogelijk dat shorters het aandeel verder onder de $ 3.00 shorten, daarom hanteren wij ook een ruime koopvisie. Let wel op dat bodempikkers ook vaak een mooie rit missen omdat zij het onderste uit de kan willen halen.
